Stay in Venice overnight, preferably 2 nights minimum. The best way to experience Venice is when the day-trippers (or cruisers) have left!
Don’t go in the peak of Summer, the heat and crowds are not pleasant.
Get away from the major tourist zones & explore the hidden neighborhoods of Venice.
Taking a gondola ride is a must. But select one away from the tourist zones, and negotiate with the gondolier. You will get a much better price and quality ride.
Don’t even bother with a map. You will get lost regardless, so embrace it. You can only go so far on an island!
